A biology laboratory on the seafloor ArchiMer
Le Bris, Nadine.
Although occupied submersibles have played an essential role in the discovery and study of hydrothermal ecosystems at depths ranging from 1500–3500 m, operational constraints at great depths have meant that the wide chemical and thermal diversity of the hydrothermal environments have long remained poorly defi ned. In the last decade, use of remotely operated vehicles (ROVs) to substantially extend dive time and development of a new set of dedicated instruments have greatly expanded our capacity to characterize seafl oor hydrothermal habitats at the interface between hydrothermal fl uid and seawater. In particular, major breakthroughs in the fi eld of in situ chemical sensing and highpressure experimentation have led to a much better understanding of the...

The ocean plays a crucial role in the functioning of the Earth System and in the provision of vital goods and services. The United Nations (UN) declared 2021–2030 as the UN Decade of Ocean Science for Sustainable Development. The Roadmap for the Ocean Decade aims to achieve six critical societal outcomes (SOs) by 2030, through the pursuit of four objectives (Os). It specifically recognizes the scarcity of biological data for deep-sea biomes, and challenges the global scientific community to conduct research to advance understanding of deep-sea ecosystems to inform sustainable management. In this paper, we map four key scientific questions identified by the academic community to the Ocean Decade SOs: (i) What is the diversity of life in the deep ocean? (ii)...

Benthic copepod communities associated with tubeworm and mussel aggregations on the East Pacific Rise ArchiMer
Gollner, S; Zekely, J; Van Dover, C; Govenar, B; Le Bris, Nadine; Nemeschkal, H; Bright, M.
Variability in the structure of the meiobenthic copepod communities was studied among three vent sites on the East Pacific Rise. One of the sites was dominated by the mussel Bathymodiolus thermophilus, and the other two of the sites were dominated by the tubeworm Riftia pachyptila. In addition to the differences in the dominant megafauna, the maximum temperatures were much lower at the mussel-dominated sites, and the temperature to sulfide ratio was different between the tubeworm-dominated sites. A total of 22 vent endemic siphonostomatoid copepod species and 5 harpacticoid copepod species were identified among the three sites. Copepod abundance was low at all sites (< 1 - 31 ind. 10 cm(-2)). Species richness ranged from 6 - 14, Shannon-Wiener diversity...

Co-occurring nematodes and bacteria in submarine canyon sediments ArchiMer
Rzeznik-orignac, Jadwiga; Puisay, Antoine; Derelle, Evelyne; Peru, Erwan; Le Bris, Nadine; Galand, Pierre E..
In submarine canyon sediments, bacteria and nematodes dominate the benthic biomass and play a key role in nutrient cycling and energy transfer. The diversity of these communities remains, however, poorly studied. This work aims at describing the composition of bacteria and nematode communities in the Lacaze-Duthiers submarine canyon in the north-western Mediterranean Sea. We targeted three sediment depths for two consecutive years and investigated the communities using nuclear markers (18S rRNA and 16S rRNA genes). High throughput sequencing combined to maximal information coefficient (MIC) statistical analysis allowed us to identify, for the first time, at the same small scale, the community structures and the co-occurrence of nematodes and bacteria...

Dissolved and particulate metals (Fe, Zn, Cu, Cd, Pb) in two habitats from an active hydrothermal field on the EPR at 13 degrees N ArchiMer
Sarradin, Pierre-marie; Lannuzel, D; Waeles, M; Crassous, Philippe; Le Bris, Nadine; Caprais, Jean-claude; Fouquet, Yves; Fabri, Marie-claire; Riso, R.
The distribution of Fe, Cu, Zn, Pb, Cd between the dissolved (<2 mu m) and the particulate (>2 mu m) fractions was measured after in-situ filtration in two hydrothermal habitats. The total metal concentration ranges exhibit a clear enrichment compared with the seawater concentration, accounting for the hydrothermal input for all the metals considered. Iron is the predominant metal (5-50 mu M) followed by Zn and Cu. Cd and Pb are present at the nM level. At the scale studied, the behavior of temperature, pH and dissolved iron is semi-conservative whereas the other dissolved and particulate metals are characterized by non-conservative patterns. The metal enrichment of the >2 mu m fraction results from the settlement and accumulation of particulate...

Distribution of bacteria and associated minerals in the gill chamber of the vent shrimp Rimicaris exoculata and related biogeochemical processes ArchiMer
Zbinden, M; Le Bris, Nadine; Gaill, F; Compere, P.
The shrimp Rimicaris exoculata dominates the megafauna of some Mid-Atlantic Ridge hydrothermal vent fields. This species harbours a rich bacterial epibiosis inside its gill chamber. At the 'Rainbow' vent site (36degrees 14.0'N), the epibionts are associated with iron oxide deposits. Investigation of both bacteria and minerals by scanning electron microscopy (SEM) and X-ray microanalysis (EDX) revealed 3 distinct compartments in the gill chamber: (1) the lower pre-branchial chamber, housing bacteria but devoid of minerals; (2) the 'true' branchial chamber, containing the gills and devoid of both bacteria and minerals; and (3) the upper pre-branchial chamber, housing the main ectosymbiotic bacterial community and associated mineral deposits. Our chemical and...

Diversity of Meiofauna from the 9 degrees 50 ' N East Pacific Rise across a Gradient of Hydrothermal Fluid Emissions ArchiMer
Gollner, Sabine; Riemer, Barbara; Arbizu, Pedro Martinez; Le Bris, Nadine; Bright, Monika.
Background: We studied the meiofauna community at deep-sea hydrothermal vents along a gradient of vent fluid emissions in the axial summit trought (AST) of the East Pacific Rise 9 degrees 50'N region. The gradient ranged from extreme high temperatures, high sulfide concentrations, and low pH at sulfide chimneys to ambient deep-sea water conditions on bare basalt. We explore meiofauna diversity and abundance, and discuss its possible underlying ecological and evolutionary processes. Methodology/Principal Findings: After sampling in five physico-chemically different habitats, the meiofauna was sorted, counted and classified. Abundances were low at all sites. A total of 52 species were identified at vent habitats. The vent community was dominated by hard...

Epifaunal community structure associated with Riftia pachyptila aggregations in chemically different hydrothermal vent habitats ArchiMer
Govenar, B; Le Bris, Nadine; Gollner, S; Glanville, J; Aperghis, A; Hourdez, S; Fisher, C.
The vestimentiferan tubeworm Riftia pachyptila (Polychaeta: Sibloglinidae) often dominates early succession stages and high productivity habitats at low-temperature hydrothermal vents on the East Pacific Rise. We collected 8 aggregations of R. pachyptila and the associated epifaunal community at 2 discrete sites of diffuse hydrothermal activity, in December 2001 and December 2002. Because of the high spatial and temporal variability of the biotic and abiotic factors related to hydrothermal vent activity, significant differences in the structure and the composition of the community were expected to occur at the scale of either 1 yr or 500 m distance between very different sites. There was no significant difference in the temperature ranges of the diffuse...

Hydrothermal circulation at ultramafic-hosted sites supports a large variety of high-and low-temperature hydrothermal vents and associated ecosystems. The discovery of abundant fossil vesicomyid and thyasirid shell accumulations at the ridge crest, approximately 2.5 km east of the active Rainbow vent field on the Mid-Atlantic Ridge (MAR, 36 degrees 13'N), increased our knowledge regarding the diversity of vent communities at slow spreading ridges. Bivalve molluscs of the family Vesicomyidae were represented by the genus Phreagena. Here we present the first record of this genus in the Atlantic Ocean. This second vesicomyid species known from the MAR, Phreagena sp., was found to be associated with a Thyasira species that is affiliated with T. southwardae (at...

Fossil evidence for serpentinization fluids fueling chemosynthetic assemblages ArchiMer
Lartaud, Franck; Little, Crispin T. S.; De Rafelis, Marc; Bayon, Germain; Dyment, Jerome; Ildefonse, Benoit; Gressier, Vincent; Fouquet, Yves; Gaill, Francoise; Le Bris, Nadine.
Among the deep-sea hydrothermal vent sites discovered in the past 30 years, Lost City on the Mid-Atlantic Ridge (MAR) is remarkable both for its alkaline fluids derived from mantle rock serpentinization and the spectacular seafloor carbonate chimneys precipitated from these fluids. Despite high concentrations of reduced chemicals in the fluids, this unique example of a serpentinite-hosted hydrothermal system currently lacks chemosynthetic assemblages dominated by large animals typical of high-temperature vent sites. Here we report abundant specimens of chemosymbiotic mussels, associated with gastropods and chemosymbiotic clams, in approximately 100 kyr old Lost City-like carbonates from the MAR close to the Rainbow site (36 degrees N). Our finding shows...

Heat-shock response and temperature resistance in the deep-sea vent shrimp Rimicaris exoculata ArchiMer
Ravaux, Juliette; Gaill, Françoise; Le Bris, Nadine; Sarradin, Pierre-marie; Jollivet, Didier; Shillito, Bruce.
The shrimp Rimicaris exoculata swarms around hydrothermal black smoker chimneys at most vent sites along the Mid-Atlantic Ridge. This species maintains close proximity to the hydrothermal fluid, where temperatures can reach 350degreesC and steep thermal and chemical gradients are expected. We performed in vivo experiments in pressurized aquaria to determine the upper thermal limit [critical thermal maximum (CTmax)] of R. exoculata and to investigate some characteristics of the shrimp stress response to heat exposure. These experiments showed that the shrimp does not tolerate sustained exposure to temperatures in the 33-37degreesC range (CTmax). A heat-inducible stress protein belonging to the hsp70 family was identified in R. exoculata, and its synthesis...

Hypotaurine and thiotaurine as indicators of sulfide exposure in bivalves and vestimentiferans from hydrothermal vents and cold seeps ArchiMer
Brand, G; Horak, R; Le Bris, Nadine; Goffredi, S; Carney, S; Govenar, B; Yancey, P.
Vesicomyid clams, vestimentiferans, and some bathymodiolin mussels from hydrothermal vents and cold seeps possess thiotrophic endosymbionts, high levels of hypotaurine and, in tissues with symbionts, thiotaurine. The latter, a product of hypotaurine and sulfide, may store and/or transport sulfide non-toxically, and the ratio to hypotaurine plus thiotauririe (Th/[H + Th]) may reflect an animal's sulfide exposure. To test this, we analyzed seep and vent animals with in situ sulfide measurements. Calyptogena kilmeri clams occur at high-sulfide seeps in Monterey Canyon, while C. (Vesicomya) pacifica clams occur at seeps with lower levels but take up and metabolize sulfide more effectively. From one seep where they co-occur, both had gill thiotaurine contents...

Influence of environmental conditions on early development of the hydrothermal vent polychaete Alvinella pompejana ArchiMer
Pradillon, F; Le Bris, Nadine; Shillito, B; Young, C; Gaill, F.
Dispersal and colonisation processes at deep-sea vents are still not fully understood, essentially because early life stages of vent species remain unknown. The polychaete worm Alvinella pompejana forms colonies on chimney walls at East Pacific Rise vent sites where the temperature can frequently exceed 20 degrees C. In vitro studies in pressure vessels showed that the early embryos tolerate temperatures in a lower range (10-14 degrees C), suggesting that they would have to escape the colony to develop. Pressure vessels offer the advantage that each parameter can be independently controlled, but they do not simulate the more complex and dynamic conditions naturally encountered at vent sites. Accordingly, in addition to incubations in pressure vessels, we...

Interactions of deep-sea vent invertebrates with their environment: The case of Rimicaris exoculata ArchiMer
Schmidt, Caroline; Le Bris, Nadine; Gaill, F.
The vent shrimp Rimicaris exoculata thrives around many hydrothermal vent sites along the Mid-Atlantic Ridge (MAR), where it aggregates into dense swarms. In contrast to hydrothermal vent fields at the East Pacific Rise (EPR), where the biomass is dominated by tubeworms, clams, and mussels, this shrimp is one of the major animal species at MAR vents. These animals are found in the dynamic mixing interface between cold oxygenated seawater and hot, reduced hydrothermal vent fluid. The adaptation of this shrimp to the hostile deep-sea hydrothermal environment and its survival strategy has been investigated since their discovery at the TAG site in the late 1980s. Rimicaris exoculata is now known to colonize black smoker complexes along the MAR in the...

Is temperature a good proxy for sulfide in hydrothermal vent habitats? ArchiMer
Le Bris, Nadine; Rodier, Philippe; Sarradin, Pierre-marie; Le Gall, Christian.
On the basis of previous studies at 9 degrees N and 13 degrees N East Pacific Rise (EPR) fields, we discuss the general assumption that temperature is a proxy for sulfide in vent fauna habitat. The main conclusion of these works is illustrated here through selected examples. In situ measurements were used to assess the variability of chemical parameters as function of temperature at the scale of fauna aggregations. Over a single vent site, aggregations of mussels or Riftia pachyptila discretely distributed around diffuse vents exhibit, in first assumption, similar sulfide-temperature correlation. In contrast, sulfide is much higher, for a given temperature, in the habitat of alvinellids than in neighboring habitats of Riftia pachyptila. Some consistency in...

Nematode communities associated with tubeworm and mussel aggregations on the East Pacific Rise ArchiMer
Zekely, J; Gollner, S; Van Dover, C; Govenar, B; Le Bris, Nadine; Nemeschkal, H; Bright, M.
The meiobenthic nematode community of three different vent sites at the East Pacific Rise was studied in order to determine whether the abundance, species richness, diversity and trophic structure were similar. The sites Tica and Riftia Field were dominated by the tubeworm, Riftia pachyptila, and the Buckfield site was dominated by the mussel Bathymodiolus thermophilus. The nematode communities of all three sites were low in abundance (< 1 up to 46 ind. 10 cm(-2)), except one sample from Tica with almost 1000 ind. 10 cm(-2). The communities at all sites consisted entirely of primary consumers, mostly deposit feeders. Species richness and Shannon-Wiener diversity indices were low and similar at both tubeworm sites and slightly but significantly higher at...
